A Universal Method for Trypsin Digestion
Initially described by Manza, et al., and further developed by Wisniewski, et al., the concept of using a spin filter for cleanup and digestion of protein samples for mass spectrometry analyses offers several advantages over more conventional approaches. In FASP (Filter-Aided Sample Prep), the spin filter forms the basis of a “proteomic reactor” for sample preparation. This proteomic reactor is used to deplete SDS, salts, and other interfering substances from the protein sample prior to alkylation and trypsin digestion. Following digestion, peptides are eluted as a concentrated solution. High molecular weight interferences such as DNA remain trapped by the filter, so they stay out of the peptide solution filtrate.
